摘    要:以槭树"秋天火焰"变色期秋季叶片为试材,采用改良CTAB法提取叶片总RNA,利用GenBank中相关物种的UFGT基因序列设计特异引物,进行RT-PCR扩增并对RT-PCR反应体系中的引物、退火温度和循环次数进行优化,建立了槭树叶片UFGT基因的RT-PCR扩增反应体系。

Abstract:The maple cultivar 'Autumn Blaze' was used as the experiment material in the article.Total RNA was extracted from the autumn leaves of maple tree by improved CTAB method.Specific primers were designed according to the conservative regions of UFGT gene from some related species listed in the GenBank database.The RT-PCR amplification was carried out.The specific primers,annealing temperature and cycle number were optimized.An optimized,effective and specific RT-PCR system for amplification of UFGT gene was established.
