刺槐花粉生活力测定方法及贮藏特性

摘    要:【目的】探索刺槐花粉生活力测定方法及其贮藏特性,为刺槐生产及育种提供参考依据。【方法】以刺槐花粉为试材,使用培养基萌发试验法测定其生活力,并研究不同浓度Ca2+、不同贮藏条件及时间对刺槐花粉萌发的影响。【结果】刺槐花粉在琼脂8.0 g/L+蔗糖20.0 mg/L+硼酸2.0 mg/L的培养基中萌发率高达30.25%;适当浓度的Ca2+能促进花粉萌发和花粉管伸长,当Ca2+浓度为1.6×10-4mg/L时,花粉萌发率高达40.25%,花粉管伸长最快;适宜刺槐花粉贮藏的条件依次为-80℃、4℃、-23℃和常温;花粉在-80℃贮藏32d后,萌发率仍达15.30%。【结论】适合刺槐花粉萌发的培养基为:琼脂8.0 g/L+蔗糖20.0 mg/L+硼酸2.0 mg/L;培养基中添加低浓度外源钙能促进刺槐花粉萌发和花粉管生长;于超低温真空冷冻条件下贮藏的刺槐花粉保持活力时间更长。

Abstract:【Objective】The assay method of pollen viability and its storage characteristics were explored to provide references for the production and breeding of Robinia pseudoacacia. 【Method】The pollen viability of Robinia pseudoacacia was detected by medium germination test. Meanwhile the effects of different Ca2+concentrations,different storage conditions and time on the Robinia pseudoacacia pollen germination were studied. 【Result】The germination rate of Robinia pseudoacacia pollen was 30.25% under 8.0 g/L agar + 20.0 mg/L sucrose + 2.0 mg/L boric acid. The proper concentration of Ca2+affected the speed of pollen germination and growth. When the concentration of Ca2+was 1.6×10-4mg/L,the pollen germination rate was 40.25%. The suitable storable temperature was-80 ℃,4 ℃,-23 ℃ and ambient temperature in turn. At-80 ℃,the germination rate of pollen was 15.30% when it was preserved for 32 days. 【Conclusion】The results showed that the most suitable medium of the Robinia pseudoacacia pollen germination was 8.0 g/L agar+20.0 mg/L sucrose+2.0 mg/L boric acid. The medium supplemented with low concentration of exogenous calcium promoted the Robinia pseudoacacia pollen germination and pollen tube growth. And Robinia pseudoacacia pollen stored in the cryogenic conditions remained longer vigor.
