山葡萄几丁质酶基因VCH3的克隆及植物表达载体的构建

摘    要:利用RT—PCR方法克隆了山葡萄几丁质酶基因VCH3的全长cDNA片段,并将该基因构建到双元载体pBI121质粒35S启动子下游,形成重组质粒pBIVCH3。通过冻融法将该重组质粒转化到根癌农杆菌LBA4404中,获得了植物表达载体。

Abstract: The full-length cDNA fragment of Vitis amurensis chitinase gene VCH3 was cloned by RT-PCR, then it was constructed to the downstream of 35S promoter in the binary vector pBI121, which formed the recombinant plasmid pBIVCH3. The plasmid pBIVCH3 was mobilized into Agrobacterium tumefaciens strain LBA4404 by freeze-melting methods, and a plant expression vector was thereby obtained.
