观赏羽扇豆离体培养生根关联酶活性及可溶性蛋白含量变化规律研究*

摘    要: 对观赏羽扇豆尖塔和奖品离体培养生根过程中叶片过氧化物酶(POD)、吲哚乙酸氧化酶(IAAO)、多酚氧化酶(PPO)3种氧化酶活性和可溶性蛋白含量进行了动态跟踪分析。结果表明:尖塔和奖品的POD活性均出现两个高峰,第1个高峰出现的时间一致,在培养后第5天;第2个高峰,奖品出现在第15天,比尖塔早5d。IAAO活性和PPO活性发展趋势相反。培养后,两个品种的IAAO活性呈现“升高〖FY(*2〗〖FY)〗降低”的趋势;PPO活性呈现“降低〖FY(*2〗〖FY)〗升高”的趋势。可溶性蛋白总体呈现波动下降趋势。研究还认为观赏羽扇豆离体培养生根过程可分为根原基诱导期、不定根表达期和不定根伸长期3个时期。根原基诱导期,POD和IAAO活性升高有利于根原基诱导。不定根表达期和伸长期,POD和IAAO活性下降,PPO活性和可溶蛋白含量维持在较高水平,有利于不定根的生长和发展。

Abstract:The purpose of this paper is to study the activity of peroxidase (POD), indoleacetic acid oxidase (IAAO) and polyphenol oxidase (PPO), as well as, the content of soluble protein of the leaves of two ornamental Lupins varieties, Minaretie and Russell Prize, in vitro. The result showed that POD activity of two species had two peaks. POD activity increased and reached their first peak at the 5th day, and then they decreased slightly following an increasing at the 10thday till reached Russell Prize′s second peak at the 15th day, Minaretie′s at 20th day. The activity of IAAO and PPO had opposite trends. IAAO activity showed “increasing declining” trend, while PPO activity showed “declining increasing” trend of two species. The content of soluble protein was declined as a whole. The study also considered that rooting processes could be divided into three phases: root primordial inducing, adventitious root expressing and adventitious root elongating. During the phase of root primordial inducing, POD and IAAO activity was increasing to induce root primordial. POD and IAAO activities were declining, as well as PPO activity and the content of soluble protein at a higher level was conducive to root growth and development.
