西伯利亚花楸组织培养关键技术的研究

摘    要:通过不同消毒试剂及培养基的选择,对西伯利亚花楸组织培养过程中的关键技术进行了研究,得到以下结论:①用70%~75%乙醇和漂白粉饱和溶液相结合的方法可以在保持低杀伤率的基础上有效地降低污染率;②分化效果最明显的培养基是MS+BA(1.0~2.0)+NAA(0.1~0.5)+2%蔗糖,加入BA效果要明显好于KT;③继代增殖最好的培养基为MS+BA0.5+NAA0.05+2%蔗糖,月增殖系数为3.0;④H+NAA(0.5~1)+2%蔗糖和1/2MS+IBA(0.5~1)+2%蔗糖培养基均可以有效地促进生根率(分别为80.7%和82.2%)和幼苗生长(平均苗高分别为4.5和3.8mm)。

Abstract:Key technical points of tissue culture of Sorbus sibirica with different kinds of disinfectant and medium are studied in this paper.Conclusion:①Using of 70%-75% Ethanol and Chlorinated Lime saturated solution can decrease the infection rate with lower killing rate effectively;②The best differentiation medium in our study is MS + BA(1.0-2.0)+ NAA(0.1-0.5)+ 2% sucrose,using BA can improve the effect better than KT;③The best subculture multiplication medium in our study is MS + BA 0.5 + NAA 0.05 + 2% sucrose,monthly proliferation coefficient is 3.0;④Mediums with H + NAA(0.5-1)+ 2% sucrose and 1/2MS + IBA(0.5-1)+ 2% sucrose can both increase the rooting rate(80.7% and 82.2%) and the growth of seedlings(4.5 and 3.8mm).
