摘    要:
以‘太平红’牡丹为材料,研究了不同灭菌方式、抗褐化物质、激素配比、培养基、生根培养条件对牡丹组织培养的影响。结果表明:以0.1%升汞液消毒8 min,再用7%次氯酸钠溶液消毒8 min灭菌效果最佳,鳞芽、土芽和种子的污染率分别为0、4.5%和22.0%;在改良MS培养基中联合添加PVPP 0.5 g/L和AC 0.5 g/L可有效减少牡丹外植体的褐化;改良MS+BA 1.0 mg/L+NAA 0.2 mg/L+GA 0.3 mg/L+CH 0.5 g/L+PVPP0.5 g/L+AC 0.5 g/L适宜牡丹鳞芽的诱导;MS+BA 1.5 mg/L+NAA 0.2 mg/L+PVPP 1.0g/L适宜于牡丹芽的增殖;1/2MS+IBA 5 mg/L+AC 0.5 g/L适用于牡丹试管苗生根。

Abstract:
The peony cultivar ’Taipinghong’ was used as a test material,and tissue culture of peony was studied under using different sterilization methods,anti-browning materials,hormone recipes, media and rooting conditions.The result showed that the effect of sterilizing with 0.1%mercuric chloride solution for 8 min and then with 7%sodium hypochlorite solution for 8 min was the best,in which the contamination rate was zero for the scaly buds,4.5%for the underground buds and 22.0% for the seeds;Adding 0.5 g/L PVPP and 0.5 g/L AC in modified MS medium could effectively mitigate browning of the peony explants;Modified MS + 1.0 mg/L BA + 0.2 mg/L NAA + 0.3 mg/L GA + 0.5 g/L CH + 0.5 g/L PVPP+0.5 g/L AC was suitable for the scaly bud induction;MS + 1.5 mg/L BA+0.2 mg/L NAA+ 1.0 g/L PVPP was suitable for the peony bud propagation;And 1/2 MS+ 5.0 mg/L IBA+ 0.5 g/L AC was suitable for the test-tube seedling rooting.
