罂粟BBE基因的生物信息学分析

摘    要:
为了对罂粟BBE基因核酸及编码的蛋白质的性质进行预测分析,并构建罂粟BBE基因的三维结构模型。运用生物信息学方法对罂粟BBE基因的理化性质、亲/疏水性、信号肽、亚细胞定位、二级结构等进行预测,同时采用同源建模的方法对罂粟BBE基因的三维结构进行模拟,并将得到的模型用能量最小化原则优化,最后用Ramachandran Plot对模型进行评估。结果表明:罂粟BBE基因全长为5073 bp,编码一段长为535个氨基酸的蛋白质。BBE为富含Gly、Ser和Leu的酸性稳定类亲水蛋白,定位于液泡中,N-末端含有23个氨基酸的信号肽,α螺旋和无规则卷曲是罂粟BBE的主要二级结构元件。通过同源建模获得了合理的罂粟BBE三维结构模型。本研究为采用生物工程技术提高小檗碱的含量、开发小檗碱类抗菌药物奠定理论基础。

Abstract:
This research aimed to analyze the structure and characters of BBE nucleic acid and protein in Papaver Somniferum L.and construct the three-dimensional structure of BBE.The author used tools of bioinformatics to analyze and predict BBE in the following aspects:biochemical function,hydrophilicity,signal peptide,subcellular localization,secondary and tertiary structure of protein and so on.At the time,construct the three-dimensional structure of BBE by comparative modeling using Discovery Studio,the model was refined by energy minimization,and assessment by Ramachandran Plot.The results showed that the full length of BBE gene and protein in Papaver somniferum L.was 5073 bp and 535 aa,respectively,BBE,which was located in the vacuole,and a stable acidic protein and rich in Gly,Ser,Leu contained a signal peptide of 23 amino acids in the N terminal.α-helix and random coil were primary secondary structural components of BBE. The author also obtained a reasonable three-dimensional structure of BBE in Papaver somniferum L.by homology modeling.The research would helpful to improve the production of berberine by genetic engineering means and develop new antibiotics.
