邓恩桉的组织培养技术研究

摘    要:
以邓恩桉(Eucalyptus dunnii)无菌苗的下胚轴和半木质化的带芽茎段为外植体,诱导芽的分化及丛生芽产生,对培养基中不同生长素和细胞分裂素的用量和配比进行了探讨,确定了邓恩桉组织培养的最适培养条件。以MS 6-BA0.2mg/L NAA0.1mg/L诱导外植体芽的分化效果最佳,产生的丛生芽易于分切,便于进行继代培养,其幼苗生长旺盛,月平均增高5mm。IBA可促进邓恩桉组培苗生根,在1/2MS培养基中添加0.2mg/L IBA,生根效果最佳。

Abstract:
Experiments were carried out to investigate the effects of different auxin and cytokinin and their combinations on the differentiation of hypocotyls from aseptic seedlings and segments with axillary buds of stems in Eucalytus dunnii, and the optimal culture conditions were determined. The results showed that MS with 6-BA 0.20 mg/L and NAA 0.1 mg/L is the most proper medium for bud differentiation and subculture, and the sprouts resulted from bud cluster was easy to be cut apart. Even more the sprouts grew bloomily and reached 5 mm in one month. The other results showed that IBA promoted Eucalyptus dunnii to strike roots and the best medium for rootage is 1/2 MS supplemented with 0.20 mg/L IBA.
