牛角瓜花粉萌发及花粉管生长研究

摘    要:运用方差分析、多重比较、正交实验和极差分析方法,对牛角瓜花粉的萌发和花粉管生长进行研究,结果表明,培养基内硼酸、硝酸钙、蔗糖、硫酸镁在一定浓度范围内,对花粉萌发及花粉管生长起促进作用,但浓度过高或过低时,则有抑制作用;钾对花粉萌发及花粉管生长影响不显著;蔗糖和硝酸钙对牛角瓜花粉萌发有显著影响,蔗糖和硫酸镁对牛角瓜花粉管生长有显著影响;牛角瓜最适花粉液体培养基为:15%蔗糖+350mg/L Ca(NO_3)2·4H_2O+60 mg/L H_3BO_3+150 mg/L MgSO_4·7H_2O;在30~35℃温度条件下,牛角瓜花粉块萌发和花粉管生长最好。

Study on pollen germination and pollen tube growth of Calotropis gigantea
Abstract:Pollen germination and pollen tube growth of Calotropis gigantea was tested by using One-Way ANOVA, multiple comparison, an orthogonal experiment and range analysis The results indicated that the pollen germination and pollen tube growth would be enhanced when the concentration of borate, calcium nitrate, magnesium sulphate and sucrose of the culture medium were within a certain range. When the concentration is too high or low, the pollen germination and pollen tube growth would be inhibited. Potassium has little impact on pollen tube growth and pollen germination. Sucrose and calcium nitrate have significant effect on pollen germination, sucrose and magnesium sulphate have significant effect on pollen tube growth. The optimum culture medium and environment conditions which support the germination and growth are 15% sucrose+350 mg/L Ca (NO3)2?4H2O+60 mg/L H3BO3+150 mg/L MgSO4?7H2O. The pollen germination and the growth of pollen tube will be better under 30~35℃.
