高产淀粉酶芽孢杆菌菌株的筛选

摘    要:[目的]筛选高产淀粉酶的芽孢杆菌菌株。[方法]选用10株芽孢杆菌,采用染色法初选和3,5-二硝基水杨酸显色法复选对其产淀粉酶的能力进行测定,筛选高产淀粉酶的芽孢杆菌菌株。[结果]10株芽孢杆菌菌株中有9株具有产淀粉酶能力,其中编号为Pab03、Pab02的2株枯草芽孢杆菌产酶活性最高,染色圈直径/菌落直径分别为2.284、1.961;在未作发酵条件优化的前提下,其酶活力分别达到(31.331±0.985)、(21.521±1.390)U,在作为新型的消化酶饲料添加剂方面具有广阔的应用前景。而菌株凝结芽孢杆菌PSAF1和枯草芽孢杆菌PJK01所产淀粉酶活力最低,分别为(0.366±0.022)、(0.704±0.089)U。[结论]成功筛选了2株高产淀粉酶的枯草芽孢杆菌菌株,分别为Pab03和Pab02。

Abstract:[Objective] The research on the screening of Bacillus strain with the ability in high-yielding amylase was conducted.[Method] The capacity of 10 Bacillus strains producing amylase was primarily tested with the staining method and finally determined with the chromogenic method of 3,5-dinitro-salicylic acid for the screening of the Bacillus strain with the ability in high-yielding amylase.
