海南霉素的遗传毒性研究及其致癌性预测:2.海南霉素体内致突变性研究

摘    要:海南霉素是我国研制成功的第一个聚醚类抗生素。在已对其进行体外致突变性检测的基础上,本文选择了2种体内试验方法,对其体内致突变性作了进一步研究。1.体内骨髓细胞染色体畸变分析;根据海南霉素的小鼠急性毒性试验结果,设立以下剂量组:0.13、0.64和1.27mg/kg,经口接毒,每日一次,连续5日,同时设溶剂和阳性对照组。试验结果经t-检验为阴性;2.体内骨髓微核试验,根据海南霉素的小鼠急性毒性试验结果,设以下剂量组:0.64、1.27和2.54mg/kg,采用Heddle(1984)提出的4次接毒和24h采样的方案,同时设溶剂和阳性对照组。试验结果经t-验为阴性。本研究结果与以往体外致突变试验结果相符,海南霉素不具致突变作用。

Abstract:Hainanmycin is the first anticoccidial polyether antibiotic developed in China.Basing on the result of the previous mutagenic tesis in vitro, 2 kinds of test in vivo had been selected for further studies on mutagenicity of hainanmycin.1. Bone marrow cytogenetics in mice according to per os LD50 of hainanmycin inmice, dose levels of the drug were as follows: 0.13,0.64 and 1.27 mg/kg. Hainanmy-cin Was administered to the mice by oral route, once a day, continously for 5 succes-sive days. Positive and negative controls were run consurrently with the dosing groups.The results of experiment examined by t-test were negative.2.Bone marrow micronucleus test in mice According to per os LD50 of hainanmycinin micel dose levels of the drug were as follows: 0.64,1.27 and 2.54 mg/kg. Thetest was performed by the Heddle's metliod(1984) reported previously. The results ofexperiment examined by t-test were negatiye.The results of these two in vivo studies were uniformly negative,and consistentwith previous in vitro mutagenic tests. The results of investigations indicate haimanmy-cin is non-genotoxic.
