四倍体刺槐无性系组织培养技术的研究

摘    要:通过茎段离体培养建立了四倍体刺槐无性系的微体繁殖体系。结果表明 :基本培养基为MS或WPM培养基。BA、NAA影响芽的增殖生长 ,在一定的范围内 ,BA对芽的增殖影响比NAA大 ,而NAA对芽高的影响比BA大 ,二者的比例对芽的增殖生长也有影响 ,芽增殖生长的最适BA和NAA组合应为BA0 5mg/L +NAA0 1mg L。四倍体刺槐无性系生根的最适生长调节物质配比为NAA0 2 5mg L +IBA0 4mg L。通过植株叶解剖观察 ,进一步证明了分步练苗能提高移栽成活率。

STUDY ON TISSUE CULTURE OF TETRAPLOID BLACK LOCUST CLONE
Abstract:A micro propagation system for tetraploid black locust( Robinia pseudoacacia ) was established by using its shoots. The optimum medium for shoot multiplication and growth was MS or WPM medium. Both BA and NAA had effects on shoot multiplication and growth. However, BA had more effect on multiplication and NAA had more effect on height of growth. At a certain concentration range, the multiplication and growth of shoots was influenced by not only the concentration but also by the ratio of BA and NAA.The best medium for multiplication and growth was MS+BA0 5 mg/L+NAA0 1mg/L. The best medium for rooting was 1/2 MS+IBA0 4mg/L +NAA0 25 mg/L. It had been proved through leaf anatomical observation that the survival percentage could be increased by means of the plantlets acclimatization.
