两种不同冷冻方法对犬冷冻精液品质的影响

本试验以德国牧羊犬的精液为材料,采用液氮熏蒸法和细管炸熏法两种冷冻方式进行细管冷冻试验,用精子活力、顶体完整率和畸形率,作为判定精子冷冻品质的指标,确定两种冷冻方法对犬冷冻精液效果的影响。结果表明:(1)液氮熏蒸法冷冻后的精子活力为0.30±0.12,细管炸熏法冷冻后的精子活力0.38±0.08,细管炸熏法优于液氮熏蒸法,且差异显著(P<0.05);(2)液氮熏蒸法冷冻后精子顶体完整率42.20±3.51,畸形率25.60±7.03,细管炸熏法冷冻后精子顶体完整率43.50±4.64,畸形率24.50±5.94,细管炸熏法稍优于液氮熏蒸法,差异不显著(P>0.05)。

Influence of Two Frozen Methods on Quality of Frozen Sperm

The semen freezing was conducted to improve the quality of German shepherd dog canine frozen semen.Straws frozen experiment was processed by liquid nitrogen vapour method and tubule frying in liquid nitrogen.Then post-thaw semen motility,the percentage of sperms with intact acrosome and the percentage of abnormality were estimated the quality of frozen semen.The results showed:(1) The sperm motility of liquid nitrogen vapour method was 0.30±0.12,the sperm motility of tubule frying in liquid nitrogen was 0.38±0.08.Tubule frying in liquid nitrogen had an advantage over the liquid nitrogen vapour method(P<0.05).(2) The percentage of sperms with intact acrosome by liquid nitrogen vapour method was 42.20±3.51 and the percentage of abnormality was 25.60±7.03,the percentage of sperms with intact acrosome by tubule frying in liquid nitrogen was 43.50±4.64 and the percentage of abnormality was 24.50±5.94.Tubule frying in liquid nitrogen was better than the liquid nitrogen vapour method,but the difference between them was not obvious.
