绿僵菌侵染对东亚飞蝗中肠保护酶和解毒酶的影响

摘    要:为了明确绿僵菌Metarhizium anisopliae对东亚飞蝗的致病机制,探讨绿僵菌对东亚飞蝗中肠保护酶系和解毒酶系的影响。采用分光光度法测定了东亚飞蝗被绿僵菌侵染后,中肠超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)、酚氧化酶(PO)、乙酰胆碱酯酶(AchE)、全酯酶(ESTs)6种酶活力的动态变化。结果表明,中肠保护酶SOD、POD、CAT呈现先上升后下降再上升的趋势。PO则呈现先升高后下降再升高最后下降的趋势;解毒酶AChE呈现先上升后下降再上升的趋势;ESTs则呈现逐渐减弱的趋势,5 d时与对照趋于一致。上述研究结果表明,东亚飞蝗被绿僵菌侵染后,不同的酶活力变化趋势存在明显差异。本研究对于揭示绿僵菌的致病机理及寄主对真菌侵染的免疫机制有一定的指导意义。

Abstract:To study the effects of Metarhizium anisopliae infection on the activity of protective enzymes (SOD, POD, CAT, PO) and detoxification enzymes (AchE, ESTs) in the midgut of Locusta migratoria manilensis for understanding the mode of action of M. anisopliae, the activity of those enzymes in the midgut of L. migratoria manilensis was monitored at 1, 2, 3, 4, and 5 d after infected by M. anisopliae using spectrophotometry. The results indicated that the activities of SOD, POD, CAT increased firstly, then decreased, and increased again at the last phase, but that of PO showed an increasing-decreasing-increasing-decreasing tendency. AchE showed the same trend like SOD, its activity also increased firstly, then decreased, and increased finally. ESTs changed differently with others overall, its activity decreased gradually and went down to the level of control at the fifth day. These results suggested that the activities of SOD, POD, CAT, PO, AchE and ESTs changed differently after L. migratoria manilensis was infected by M. anisopliae, it will contribute to uncover the mode of action of M. anisopliae in the midgut of hosts and the immune mechanism of hosts against pathogens.
