三尖杉愈伤组织诱导及其分化的初步研究

以三尖杉茎尖、嫩叶为外植体,应用正交设计法对愈伤组织诱导及其继代分化进行了研究。结果表明,不同外植体灭菌的难易程度相差较大,叶片较茎尖容易;叶片用体积分数为75%的酒精浸泡30-45 s,经体积分数为0.1%HgCl2溶液灭菌12 min,效果最好;茎尖则用体积分数为75%酒精浸泡1 min,经体积分数为0.1%HgCl2溶液灭菌12 min,效果较好;诱导叶片、茎尖愈伤组织的最佳培养基配方为MS+NAA 1.0 mg.L-1+2,4-D 1.0 mg.L-1;愈伤组织在继代培养基上均能进行分化,其中结构紧密的茎尖愈伤组织在MS+6-BA3.0 mg.L-1培养基上继代培养能诱导分化形成不定芽。

Preliminary study on callus induction and differentiation of Cephalotaxus fortunei

With the orthogonal design,the young stem tip and tender leaf of Cephalotaxus fortunei Hook.f.were used for tissue culture and the induction and differentiation of callus.The results indicated that there was difference between sterilization of explants,the tender leaf was disinfected easily.The best sterilization procedure for the tender leaf was 0.1%HgCl2 for 12 minutes after 75% alcohol for 30-45 seconds and the best sterilization procedure for the stem tip was 0.1%HgCl2 for 12 minutes after 75% alcohol for 1 minutes.The optimum medium for callus of the tender leaf and stem tip was MS+NAA 1.0 mg·L-1+2,4-D 1.0 mg·L-1 and the callus could difgerentiate on the subculture,the callus from the stem tip which has close fabric could induce the adventitious buds on the subculture MS+6-BA 3.0 mg·L-1.
