新疆南部3个地方品种绵羊的遗传相关性分析

摘    要:
利用随机扩增多态性DNA(RAPD)技术研究了多浪羊、叶城羊和卡拉库尔羊间的亲缘关系.从40条随机引物中筛选出15条具多态性引物,其中8条多态性引物的稳定性较好,扩增产物用1%琼脂糖凝胶电泳分离,用凝胶成像系统对电泳图谱进行分析并拍照保存.将实验结果进行聚类分析,发现多浪羊与叶城羊(遗传距离0.670)、卡拉库尔羊(遗传距离0.540)的亲缘关系均较远;另外,卡拉库尔羊产双羔成年母羊与公羊的遗传距离小于绵羊种间遗传距离(遗传距离0.072),与产单羔成年母羊的遗传距离最小(遗传距离0.063).

Abstract:
The technology of Random Amplified Polymorphic DNA(RAPD) was used to detect the DNA polymorphism of Duolang,Yecheng and Karakul sheep.Forty primers were used to analyze the polymorphism of three local sheep varieties,in which the amplification results of 8 primers showed obviously polymorphous.The amplified products were detected with 1% agarose gel electrophoresis,taken in pictures and recorded with the information of Gel DOC 2000 systematically.The results indicated that the genetic relationship of Duolang and Karakul sheep was comparatively further(genetic distance 0.540),and Duolang and Yecheng sheep was comparatively further(genetic distance 0.670) too.In addition,the relationship between the female Karakul that produced pairs of lamb and male Karakul was less than the distance of species(genetic distance 0.072),and the female Karakul producing pairs of lambs and Karakul female producing single lamb was relatively small(0.063).
