曲面分析在‘雪花梨’叶片不定芽诱导培养基筛选上的应用

摘    要:以‘雪花梨’试管苗幼叶为外植体,采用正交设计和曲面分析的方法对叶片不定芽诱导培养基进行筛选。结果表明,‘雪花梨’叶片在MS+6-BA1.0mg/L+NAA0.05mg/L+葡萄糖30g/L+琼脂6g/L培养基上诱导不定芽再生芽数为2.95,高于其他处理;在MS+6-BA1.0mg/L+TDZ1.0mg/L+NAA0.05mg/L+葡萄糖30扎十琼脂6g/L培养基上再生率最高,达75%。经过曲面分析,‘雪花梨’在MS+TDZ1.0mg/L+6-BA0.5mg/L+蔗糖30g/L+琼脂6g/L的培养基上进行培养,理论再生芽数可达7.00,在MS+TDZ1.0mg/L+6-BA1.0mg/L+NAA0.05mg/L+蔗糖30只,L+琼脂6玑的培养基上进行培养,理论再生率可达100%。

Abstract:Orthogonal design and surface analysis methods on culture medium screening of Adventitous BudsInduction from Leaves in vitro of ‘Xuehua pear’ were adopted.The results showed that ‘Xuehua pear’ leavesregeneration buds were 2.95 in the MS+6 -BA 1.0 mg/L+NAA 0.05 mg/L+glucose 30 g/L+agar 6 g/L cul-ture medium,higher than other treatments.Regeneration rate was the highest,up to 75% in the MS+6 -BA1.0 mg/L+TDZ 1.0 mg/L+NAA 0.05 mg/L+glucose 30 g/L+agar 6 g/L culture medium.After the curve sur-face analysis,‘Xuehua pear’ regeneration buds were 7.00 in the MS+TDZ 1.0 mg/L+6-BA 0.5 mg/L+su-crose 30 g/L +agar 6 g/L culture medium.Regeneration rate was 100% in MS+6-BA 1.0 mg/L+TDZ 1.0 mg/L+ NAA 0.05 mg/L+glucose 30 g/L+agar 6 g/L culture medium.
