摘    要:以野败型细胞雄性不育系珍油97A与其强恢复系IR24配组,构建由210株组成的F2代极端不育群体作为恢复基因定位群体。用ISSR引物UBC835对两个亲本进行扩增,发现PCR指纹在两个亲本间具多态性,再用此引物对基因定位群体进行连锁分析表明,UBC835与水稻野败型细胞质雄性不育恢复基因紧密连涣,交换率为3.57%,转换成图距为3.58cM。经过150多对微卫星引物进行扫描,在第1、第7染色体和第

Abstract:To develop the PCR based molecular marker closely linked to the restorer gene in the wild abortive (WA) type cytoplasmic male sterility (CMS) system, the F 2 excessive sterile population consisting of 210 plants from the cross between Zhenshan 97A and the strong restorer line IR24 was used for mapping. A ISSR marker,UBC 835, was found to generate polymorphism between the two parents. Linkage analysis in the mapping population showed that UBC 835 was closely linked to the restorer gene of the CMS WA system. The exchange frequency was 3.57%,the distance between UBC 835 and the restorer gene was 3.58 cM. The same results was obtained by linkage analysis with OSR33 located on chromosome 10 in the rice genome by screening more than 150 SSR loci,however, no SSR locus was found to link with the restorer gene in the polymorphic SSR loci on the chromorsome 1 and 7. The use of the two PCR based molecular marker closely linked to the restorer gene in molecular assistant selection for restorer line was also discussed.
