北虫草超氧化物歧化酶的分离纯化及酶学特性

摘    要:研究北虫草超氧化物歧化酶(SOD)的分离纯化和部分酶学特性。结果表明,从北虫草中提取SOD的最佳工艺条件为原料与PBS(pH7.73)配比为1∶3(m∶v),提取时间为3h,纯化倍数可达到粗酶液的4.09倍。提纯后的SOD在690nm左右出现了特征吸收峰,最适温度38℃,最适pH值为7.8,大部分糖对SOD的活性没有显著影响,有机酸都有降低酶活性的作用,过氧化氢对该酶有抑制作用,北虫草SOD是以铜和锌离子为辅基,为北虫草的进一步开发和利用提供了理论依据。

Abstract:The properties of superoxide dismutase (SOD) from Cordyceps militaris were studied. The optimal isolating conditions were that the ratio of crude extract and PBS was 1∶3 (m∶v) , the pH value of PBS was 7.73, and the extraction time was 3 h. The purification multiple was 4.09 by column chromatography. The purified SOD showed characteristic peak at 690 nm. The optimum temperature and pH were 38℃ and 7.8, respectively. Lactose, sucrose, glucose and maltose had no impact on the activity of SOD. The activity of SOD was inhibited by organic acid and H2O2. SOD of C. militaris was Cu, Zn-SOD. SOD from C. militaris were isolated and purified successfully, and its partial enzymatic properties were studied, which provided important information for future applications and research.
