影响农杆菌介导的河北杨遗传转化的因素

摘    要:从预培养时间、侵染菌液浓度、侵染时间、共培养时间及添加A S(乙酰丁香酮)5个方面,以卡那霉素抗性芽频率作为衡量指标,研究了各因素对河北杨遗传转化率的影响。优化筛选的最适转化系统为:预培养2 d,农杆菌活化菌液1/2M S稀释10倍,侵染5 m in,共培养4 d。按此方法河北杨叶盘转化率可达35%。

Abstract:The effects of influencing factors were studied,such as pre-culture time,Agrobacterium concentration,infection time,co-culture time and adding AS on genetic transformation frequency of Populus hopeiensis,of which the frequency of kanamycin resistance buds was used as measure standard.The superior transformation system was as following,pre-cultured two days and then infected 5 minutes in Agrobacterium diluted ten times with 1/2MS liquid medium,co-cultured four days at last.The transformation frequency of leaf disc of Populus hopeiensis would reach to 35% with this system.
