摘    要:用水稻、玉米、棉花和油菜的干种子作材料进行了分离DNA的试验。试验结果表明,从4种作物的干种子中都能分离到质量较好的DNA,得到的DNA可以用于RAPD和SSR分析。这一方法最显著的特点是充分利用了DNA分离的原理,只保留了细胞裂解、DNA沉淀和溶解等关键步骤,而省略了二些诸如去除蛋白质及有机物的中间步骤,因而具有简便、快速等优点,特别适合于一些需要快速得到研究结果的分析,如种子纯度鉴定、分子标记辅助选择等。

Abstract:A rapid DNA isolation method was established in this study. Dry seed of rice,maize and cotton was adopted as testing material. The integrity of DNA isolated with this method was satisfactory and can meet the requirement of the analysis for RAPD and SSR. The distinct characteristic of this method is that it only preserves the steps of tissue disruption, DNA precipitation and dissolution, but discards steps such as elimination of protein and organic compound. The method is simple and rapid and it is especially suitable for analyses if the results of which need to be obtained instantly; such as the identification of seed purity and molecular marker assistant selection.
