摘    要:目的:建立小麦中杂色曲霉毒素含量的高效液相色谱检测方法。方法:样品中的杂色曲霉毒素经正己烷提取后,采用国产硅镁型吸附剂制备层析柱净化后,运用高效液相色谱法测定,色谱柱为Nova-Pak C18柱,以甲醇-水(70∶30,V/V)为流动相,流速0.7mL/min,检测波长为246nm进行检测。结果:该方法平均加标回收率为79.26%,相对标准偏差为3.84%,最低检出量为0.017μg/kg。结论:本方法简便、准确、成本低,可用于小麦中杂色曲霉毒素含量的测定。

Abstract:HPLC method for the determination of sterigmatocystin in grains was developed.Sterigmatocystin was extracted by hexane and cleaned-up using florisil column.Analysis was carried out by using Nova-Pak C18 column with an isocratic mobile phase composed of methanol-water(70∶30,V/V),flow speed 0.7 mL/m and UV detection at 246 nm.Results: The average recovery was 79.26% with the RSD of 3.84%,limit of detection was 0.017 μg/kg.Conclusion: The method is simple,accurate,cheaper,and can be used for the determination of sterigmatocystin in wheat.
