春石斛根端染色体制片技术的优化

摘    要:为优化春石斛染色体制片技术体系,以春石斛组培苗根为材料,研究根端长度、预处理方法和酸解时间对制片的影响,结果表明,春石斛根端染色体制片时取根尖长度为1.0cm,在8-羟基喹啉和饱和对二氯苯的混合液中预处理4h,用1mol/L的盐酸在常温(25℃)下解离18min时制片效果佳,染色体形态清楚,易于观察。

Abstract:In order to optimize technology system of chromosome sectioning,usethe root tips of plantlets in vitro of nibole-type Dendrobium to study the effects of root tip length,methods of pretreatment,and time of acidolysis.The result showed that clear and observable chromozomes were found when the root tips were cut into 1.0 cm,and pretreated with 8-hydroxyquinoline and p-bromonaphthalene for 4 hours,and macerated in 1 N HCl solution for 18 minutes at room temperature(25 ℃).
