摘    要:为了给长寿沙田柚的染色体核型分析、细胞遗传学研究和良种培育提供参考依据,以其幼叶为研究材料,设计4种前低渗时间和12种酶解方式处理,对制片效果的影响进行了研究。结果表明:预处理后的材料经前低渗60min后,用0.2%纤维素酶∶0.2%果胶酶(3∶1,v/v)混合酶液37℃酶解80min,可得到理想的制片效果,背景干净,染色体分散、清晰。

Abstract:In order to provide a reference for the chromosome karyotype analysis,cytogenetic research and cultivation of improved varieties of C.grandis,taking the young leaves as the study materials,the effects of the key steps to chromosome squashing were studied in the process of wall digestion hypotonic in this paper.The experiment was designed into four different early low osmosis treating times,12 different enzyme digestion treatments.The results showed that the technique was workable for pretreated samples dealing with early low osmosis for 60 min,then digested by 0.2% cellulase and 0.2% pectinase(3∶1,v/v)for 80 min.The chromosome images were satisfied with clean background,chromosomes dispersing well and clear form.
