薏米蛋白的酶水解工艺优化

摘    要:为确定蛋白酶水解薏米蛋白的最佳工艺条件,以氮溶解指数(NSI)和水解度(DH)为指标对中性蛋白酶、碱性蛋白酶、木瓜蛋白酶、胰蛋白酶进行筛选,确定碱性蛋白酶为最佳水解用酶,并通过单因素和二次回归正交旋转试验,建立了碱性蛋白酶水解薏米蛋白的数学模型。结果表明,碱性蛋白酶在底物质量浓度39.95 g/L,酶用量1 201.75 U/g,温度54.61℃,pH值7.99,反应时间4 h的条件下,水解后的氮溶解指数可达95.79%。

Abstract:In order to optimize the optimal condition of hydrolysis for coix seed protein by protease,taking nitrogen solubility index and degree of hydrolysis as the indicator,select the best hydrolysis protease from the neutrase,alcalase,papain,and trypsin.Finally,identify alcalase is the best hydrolysis protease.Based on the single factor experiment and second-order regression rotation design,established the protease hydrolysis mathematical model.The results show that the optimal condition of alcalase is:substrate concentration is 39.95 g/L,the protease dosage is 1 201.75 U/g,temperature is 54.61℃,pH is 7.99,and time is 4 h.Under the optimal conditions,nitrogen solubility index of coix seed protein hydrolyzed by protease can reach 95.79%.
