重组人溶菌酶体外抑菌活性研究

摘    要:
以购买商品化的溶菌酶标准品作为对照,通过平板扩散法检测重组人溶菌酶(rhLYZ)在体外对几株致病菌及益生菌的抑菌作用,同时用梯度稀释法检测重组人溶菌酶的MIC和MBC。分别将重组酶和商品酶做梯度稀释后,滴加到涂布有一定量菌液的MH平板,通过平板扩散法测量各组抑菌环直径大小,从而判定其抑菌活性。结果表明,重组人溶菌酶抑菌活性显著高于标准溶菌酶,低质量浓度的重组酶即可对致病菌的生长产生抑制和杀灭作用。rhLYZ对金黄色葡萄球菌、致病性大肠埃希菌较为敏感,抑菌环直径达到15mm~18mm;对多杀性巴氏杆菌抑菌环直径则达到27.5mm。提示rhLYZ对金黄色葡萄球菌、致病性大肠埃希菌及多杀性巴氏杆菌等病原菌均具有较强抑菌活性。

Abstract:
Using the flat diffusion method and comparing with a commercialized lysozyme control,recombinant human lysozyme(rhLYZ) in vitro antibacterial activities on several pathogenic bacteria and probiotic bacteria strains,were analyzed.At the same time,the MIC and MBC of the rhLYZ on different bacteria were detected by serial dilution method.Diluted rhLYZ and commercialized lysozyme were added onto MH plates which contain certain volume of bacteria.Antibacterial activities were determined by measuring the diameter of bacteriostatic ring.The results showed that the recombinant human lysozyme had notable higher antibacterial activity than the commercialized lysozyme.Lower concentration of the rhLYZ could inhibit the growth of pathogenic bacteria,especially more sensitive on Staphylococcus aureus and Escherichia coli.The diameter of antibacterial circle was 15 mm-18 mm and the highest record was 27.5 mm on Pasteurella multocida.This implied that rhLYZ had a strong bacteria inhibition activity on pathogenic bacteria like Staphylococcus aureus,Escherichia coli,Pasteurella multocida,etc.
