利用半冬性甘蓝型油菜扩大甘蓝型春油菜细胞质雄性不育系与恢复系间遗传差异的研究

摘    要:以2个半冬性甘蓝型油菜品种(系)、2个春性甘蓝型恢复系、2个春性甘蓝型不育系以及16个由2个春性恢复系与2个半冬性品种杂交选育的新春性甘蓝型恢复系为材料.利用SSR、SRAP、AFLP 3种标记对这些材料的亲缘关系进行研究,同时对新恢复系和相应的亲本恢复系与2个春性不育系间的遗传差异进行比较.3种标记的综合聚类结果表明,在相似系数为0.583处将22份材料分成4类,春性亲本恢复系Ag-5和Qu、2份两春性不育系聚为第Ⅰ类;8份新春性恢复系聚为第Ⅱ类;半冬性亲本LB和5份新春性恢复系聚为第Ⅲ类中;第Ⅳ类包括半冬性亲本305和3份新春性恢复系.16份新恢复系中931-935与2份不育系的遗传距离均小于其亲本恢复系Ag-5与2份不育系的遗传距离,帐23与不育系Ⅰ的遗传距离小于其亲本恢复系Ag-5与不育系Ⅰ的遗传距离,而其余14份新恢复系与2份不育系的遗传距离均大于其相应春性亲本恢复系与2份不育系的遗传距离,说明向春性恢复系中导入半冬性品种遗传成份能有效扩大恢复系与不育系间的遗传差异.

Abstract:The Brassica napus materials include 2 weak-winter rapeseed parents,2 spring restorer lines,2 spring male sterile lines and 16 new spring restorer lines derived from the hybrids of 2 spring restorer parents and 2 weak-winter rapeseed parents.The molecular markers of SSR,SRAP and AFLP were used to study the genetic relationship of those materials,to explore the genetic differences between the new bred lines and corresponding parental restorer lines and two spring male sterile lines.Results indicated that all the materials can be grouped into 4 classes when the similarity coefficient was 0.583.2 based on cluster analysis of three molecular markers.Spring restorer lines Ag-5,Qu and 2 spring male sterile lines were grouped into class I.Eight new spring restorer lines were grouped into class Ⅱ.Weak-winter rapeseed parent LB and 5 new spring restorer lines were grouped into class Ⅲ,and Class Ⅳ contained weak-winter rapeseed parent 305 and 3 others new spring restorer lines.Among 16 new restorer lines,the new restorer line 931-935 showed closer genetic relationship with two male sterile lines than their restorer parent Ag-5,and Tent 23 also showed closer genetic relationship with Sterile Ⅰthan its restorer parental Ag-5.While other 14 new restorer lines showed greater genetic relationship with the two male sterile lines than the corresponding spring parents.These results indicated that the genetic insertion of semi-winter varieties into spring restore lines can effectively expand the genetic difference between restorer lines and CMS lines.
