高效聚磷鞘氨醇杆菌XF-5的分离与鉴定

摘    要:采用寡营养与长时间培养方法,从活性污泥与土壤样品中分离筛选到7株高效聚磷菌。以菌株的除磷率为考察指标,确定其中的XF-5为高效聚磷菌株。对其培养条件进行优化,并进行了生理生化特性的检测与系统发育分析。16SrRNA基因序列的分析结果显示,XF-5与鞘氨醇杆菌属的菌种同源性达99%,结合生长特点与生理生化特性,初步确定该菌株为鞘氨醇杆菌(No-vosphingobiumsp.)。当合成废水中总磷质量浓度为10.0mg/L时,将菌株XF-5在28℃、pH值为6.5的条件下培养24h,除磷率可达80%以上。该菌株在污水除磷与水体富营养化的防治方面,有潜在的应用前景。

Abstract:A oligotrophic medium culture conditions was adopted to isolate and screen high phosphate accumulating bacteria from activated sludge and soil samples.After testing the phosphate accumulating rates,the strain XF-5 was chosen as a high efficient strain.In this study,physiological and biochemical characteristics and phylogenetic analysis were conducted for identification of the high efficient phosphorus removal strain.16S rRNA gene sequence analysis showed that XF-5 and other species of the genus Novosphingobium had a homology of 99%.Combined with the growth and physiological and biochemical characteristics,XF-5 was finally identified as a member of Novosphingobium sp.When cultured in synthetic wastewater with total phosphorus concentration of 10 mg/L at 28 ℃,pH value of 6.5 for 24 h,the phosphorus accumulation rate of the strain XF-5 could reach more than 80%.So the strain could be used to remove phosphorus of sewage and then used to control eutrophication.
