| 摘 要： | 将本实验室保存的ZJ2000株囊毒加生理盐水研磨后接种9～10日龄鸡胚增殖病毒，收集尿囊液透析纯化，用蛋白酶K法提取病毒基因组dsRNA。通过RT-PCR获得基因组B节段全长的eDNA。并将其克隆到PUC18-T载体中进行测序。同时，本研究对GenBank中登录的IBDV基因组B节段进行了大量分析，发现B节段的5＇-NCR存在1个基因高突变区，并且ZJ2000株的5＇-NCR可形成独特的二级结构。经过对VP1一级结构的大量分析，筛选出B节段中17个可能对IBDV毒力产生影响的候选毒力位点，并在此基础上又对这些位点氨基酸的特性进行统计分析，进一步探讨了这些位点对VP1功能影响的可能性，为深入研究IBDV基因组B节段对其毒力的影响作了有益的探索。

| Abstract: | cDNA of the full-length segment B of infectious bursal disease virus ZJ2000 strain was obtained by RT-PCR and cloned into PUC18-T for sequencing.Analysis of segment B sequences in GenBank identified a highly variable region in 5'-NCR.The 5'-NCR of ZJ2000 strain contained sequence that potentially forms a unique secondary structure.The deduced amino acids sequences of the VPl gene suggested that there were 17 possible loci in segment B that may affect virulence. |
