副溶血性弧菌增菌培养基及培养条件的改进

摘    要:对副溶血性弧菌增菌培养基及培养条件进行了改进和优化,建立一种更适用于副溶血弧菌增菌的方法。针对能够决定副溶血弧菌综合营养需求的2个最为关键的因素:混合蛋白胨、酵母浸膏以及两者交互作用,进行试验设计,结合统计学软件对结果进行直观分析和方差分析,验证最佳培养基和培养条件。直观分析表明,副溶血弧菌增殖过程中,混合蛋白胨是最主要因素,其次为酵母浸膏。方差分析表明,混合蛋白胨与酵母浸膏对副溶血弧菌生长的影响均显著。最佳培养基配方为:混合蛋白胨2%(胰蛋白胨1%、鱼蛋白胨1%)、酵母浸膏0.2%、NaCl 3.5%、pH8.6;最佳培养条件为:(36±1)℃+摇床120 r/min+2层牛皮纸封口。本优化培养基与培养条件用于副溶血弧菌增菌培养,在相同培养时间内,其增菌菌液浓度为GB/T4789.7-2008方法的(1.8±0.6)倍。

Abstract:The bacterium Vibrio parahaemolyticus enrichment medium and culture conditions were optimized by combination of two decisive factors peptone and yeast extract in the experiment designed by statistical software in order to obtain a method for enriching the bacterium.Direct analysis showed that the mixed peptone was found to be the major factor while the yeast extract was considered as the minor in the process of enrichment.The interaction between the two was ignored because of the weak performance.Variance a...
