摘    要:通过单因素及正交试验,研究了碳源、有机氮源、无机氮源对重组大肠杆菌Eschericha.coli BL21(DE3)/ pET-28a-est825 菌体生长及产酶的影响,进而优化了产酶培养基,确定最佳培养基组成为院甘油20 g/L,酵母粉20 g/L,硫酸铵5 g/L,NaCl 5 g/L,Na2HPO4窑12H2O 10.7 g/L,KH2PO4 2.7 g/L,硫酸卡那霉素50 滋g/L,pH 7.0。采用此培养基 发酵所得发酵液酶活力较优化前提高了86%。

Optimization of medium constituents for pyrethroid pesticide degradative enzyme expression by recombinant Escherichia coli in shake flask fermentation
Abstract:The medium for pyrethroid pesticide degradative enzyme expression by recombinant Escherichia coli was optimized by investigating the effcets of carbon sources, organic nitrogen sources and inorganic nitrogen sources on cell growth and expression of degradative enzyme through single facor and orthogonal experiments. The results showed that the optimal medium composition was glycerol 20 g/L, yeast extract 20 g/L, (NH4)2SO4 5 g/L, NaCl 5 g/L, Na2HPO412H2O 10.7 g/L, KH2PO4 2.7 g/L, kanamycin sulfate 50 g/L, pH 7.0. By using the optimum medium, the activity of degradative enzyme increasedby about 86% compared with the original medium.
