Abstract:An experiment was conducted to study the effects of sterilization time and inoculation methods on the bulb scale culture in vitro of Oriental Lily Sorbonne.The influences of hormone concentrations on induction,proliferation,small bulb cultivation and rooting were also studied.Results showed that the bulb scales treated with 70% alcohol for 30s followed by 0.1% HgCl2 for 15min exhibited the best sterilization effect,with a survival rate of 66.0%.The regeneration ability of external scale was the best,followed by middle scale,internal scale,external layer,middle layer,and internal layer.The induction rate of bulb scales after low temperature pretreatment(4 degrees C) could reach 94.4%.The optimal culture medium for induction was MS supplemented with 1.5mg·L-1 6-BA and 0.2mg·L-1 NAA,with an induction rate of 97.5%;that for multiplication was MS containing 2.0mg·L-1 6-BA and 0.2mg·L-1 NAA,and the multiplication coefficient reached 8.0;the optimum media for bulblet growth and rooting were MS with 1.0mg·L-1 BA and 1.0mg·L-1 NAA and MS with 0.5mg·L-1 BA and 0.05mg·L-1 NAA,on which the perimeter of the bulblets could reach 5.5cm with strong roots.
