摘    要:采用phenomenex C_(18)色谱柱(150 mm×4.6 mm,5μm),以甲醇-冰醋酸(pH=3.10)(45:55)为流动相洗脱,体积流量为0.8 mL/min,紫外检测波长为327 nm,采用外标法测定不同产地杜仲中绿原酸和芦丁的含量.建立了HPLC法测定杜仲中绿原酸和芦丁含量,并对不同产地杜仲中绿原酸和芦丁舍量进行测定.结果表明,绿原酸和芦丁分别在0.127 5~24.000 0μg(r=0.999 6),0.125 0~25.000 0 μg(r=0.999 6)线性关系良好,平均回收率分别为98.73%和97.26%,相对标准偏差为1.85%和1.32%.
Abstract:
Objective To establish an HPLC method for the determination of chlorogenic acid and rutin and use it to determine chlorogenic acid and rutin in Eucommia ulmoides Oliv produced in different places.Methods A phenomenex C_(18) column(150 mm×4.6 mm,5μm)was used,with a mobile phase of methanol-acetic acid(pH=3.10)(45:55)for elution;the flow rate was 0.8 mL/min;the detection wavelength was at 327 nm;and the contents of chlorogenic acid and rutin were determined by the external standard method.Results The calibration curves for chlorogenic acid and rutin were linear between 0.127 5 and24.000 0μg and between 0.125 0 and 25.000 0 μg(r=0.999 6 and 0.999 6),respectively.Their average recoveries were 98.73%and 97.26%and RSD were 1.85%and 1.32%,respectively.Conclusion The method is simple,rapid,sensitive and reproducible,and thus provides a scientific means for establishing the quality standard for Eucommia ulmoides Oliv collected from different places.
