摘    要:试验选用果皮无蜡粉西瓜品系‘W1-1’为母本,果皮有蜡粉西瓜品系‘1061’为父本,配制杂交组合获得F2代群体,分析果皮蜡粉性状遗传规律发现,西瓜果皮蜡粉性状受一对显性基因控制。利用群体分离分析(BSA)法筛选得到西瓜果皮有蜡粉基因位于第8号染色体,以双亲材料基因组重测序为基础,在定位区域开发30对引物,其中19对引物具有多态性。利用19对引物标记F2代群体,构建分子遗传图谱,该图谱全长230.29 cM,定位到一个距离为2.78 cM控制西瓜果皮蜡粉的位点,两个与该位点紧密连锁的CAPS标记,分别为w8-6149和w8-0794。利用100份西瓜自然群体材料分析2个与西瓜果皮有蜡粉紧密连锁标记有效性,分子数据与田间数据吻合率分别为79.0%和72.0%。

Study on mapping gene related to wax powder in watermelon

Abstract:The test selected the pericarp non-waxy watermelon strain 'W1-1' as the female parent,the pericarp with waxy watermelon strain '1061' as the male parent to prepare the hybrid combination was prepared to obtain the F2 generation population. By analyzing the genetic laws of the pericarp wax powder traits, it was found that the characters of watermelon pericarp wax powder were controlled by a pair of dominant genes. Using the group separation analysis(BSA) method, the wax powder gene of watermelon pericarp was located on chromosome 8. Based on the resequencing of the genome of parental materials, 30 pairs of primers were developed in the positioning area, of which 19 pairs were polymorphized. Using these 19 pairs of primers to mark the F2 generation population, a molecular genetic map was constructed, and finally a site with a distance of 2.78 cM to control the wax powder of watermelon peel was obtained, and two closely linked markers with the site were obtained. They were w8-6149 and w8-0794, respectively, and the map was 230.29 cM in length. Using 100 natural population materials of watermelon to effectively analyzed the two markers of close linkage of wax powder with watermelon pericarp, the coincidence of molecular data and field data was 79.0% and72.0%, respectively.
