新疆甜瓜枯萎病、根腐病病原菌鉴定

摘    要:目的]采用形态学观察和分子鉴定方法对甜瓜病原菌新疆株(前人鉴定为枯萎病病原菌Fusarium oxysporum(Schi)f.sp.melonis Snyder et Hansen)、枯萎病病原菌辽宁株和根腐病病原菌(Fusarium solani (Mart.) Sacc.)进行研究,确定病原菌新疆株的种类.结果]对病原菌新疆株进行形态学观察发现其有蓝色色素,维管束变褐不向上发展.此后又对三种病原菌进行了大亚基rDNA-D1/D2区序列分析、rDNA-ITS区序列分析,两者分析结果表明病原菌新疆株与根腐病病原菌同源性高.结论]分子鉴定与形态学鉴定结果一致,表明病原菌新疆株为腐皮镰孢菌(Fusarium solani (Mart.) Sacc.)而非尖镰孢菌(Fusarium axysporum(Schi.)f.sp.melonis Snyder et Hansen),是根腐病病原菌,与前人鉴定结果不同.

Abstract:【Objective and Method】The pathogen of melon Xinjiang strain(Pioneer identified it as Fusarium axysporum(Schi.) f.sp.melonis Snyder et Hansen),liaoning strain and root rot(Fusarium solani(Mart.) Sacc.)were identified on the basis of morphological characters and molecular identification inorder to classify the pathogen of Xinjiang strain.【Result】Morphogenesis observation of Xinjiang strain showed that this pathogen can accumulate blue pigment and brown vascular bundle confined to limited domain.This study analysis the rDNA-D1/D2 and rDNA-ITS sequence of large ribosome subunit which belong to Xinjiang strain,Liaoning strain and root rot disease pathogen,results showed that there is high homologous characters between Xinjiang strain and root rot disease.【Conclusion】The reuslt of molecular identification showed no difference with the results of morphology identification,indicating that the pathogen of Xinjiang strain belongs to Fusarium solani(Mart.)Sacc.not Fusarium axysporum(Schi.) f.sp.melonis Snyder et Hansen,is pathogen of root rot,different from the results of which pioneer identified.
