摘    要:以香灰菌HYP-GT01为试材,通过调节培养基成分、pH和光照强度,研究不同培养条件下菌丝生长及黑色素产量差异,以期发现影响香灰菌菌丝生长和黑色素产生的主要因素。结果表明:香灰菌在完全培养基上菌丝生长速度最快,菌丝致密边缘整齐,产生黑色素且产量最高,达0.42g·L-1;在低氮培养基上菌丝生长较慢,但菌丝浓密,可产生黑色素,达0.34g·L-1。碳源不足菌丝纤细稀疏,且不产黑色素;高无机盐含量和添加吐温20会对菌丝生长产生抑制,可产少量黑色素。在一定pH范围内酸碱度对该菌株菌丝生长及黑色素的产生影响不大。不同光照条件不仅影响菌丝生长速度,还影响黑色素的产生。

Abstract:Using Hypoxylon HYP-GT01 as test material,by adjusting the composition of culture medium,pH and light intensity,the morphological characteristics,growth rate and melanin yield of hyphal under different culture conditions were analyzed in order to find out the main factors affecting the hyphal growth of Hypoxylon.The results showed that the growth rate of the hyphae was the fastest in the culture medium.The dense edge of the hyphae was neat.The melanin was produced,and the yield was 0.42 g·L-1,was the highest.The hypha growth was slow in the low-nitrogen medium,but the hyphae were thick,and the melanin could be produced up to 0.34 g·L-1.The carbon source wasn′t enough for the filament to be thin and sparse,and it didn′t produce melanin.High inorganic salt content and the addition of Tween 20 could inhibit mycelium growth and produce a small amount of melanin.The pH level in a certain range had little effect on the growth and melanin.Different light conditions were not only affect the growth rate of mycelium,but also the production of melanin.
