响应面方法优化仙人掌黄酮类物质提取工艺条件的研究

摘    要:
[目的]研究乙醇溶剂提取仙人掌中黄酮类物质的最佳工艺条件。[方法]以仙人掌干粉为试材,以乙醇溶液为提取溶剂,采用响应面设计方法,对影响黄酮提取效果的4个因素乙醇浓度、提取温度、提取时间和液料比先后进行部分因子试验和中心组合设计试验,并建立数学模型,研究这些因素对黄酮提取率的影响。[结果]部分因子试验结果表明,乙醇浓度和料液比是影响仙人掌黄酮提取效果的主要因子;中心组合设计试验建立的黄酮提取率(Y)与乙醇浓度(A)、液料比(B)间的数学模型为Y=-13.5872+0.2961A+0.1203B-0.00174A2-0.0030B(2R2=0.948);最佳的提取工艺条件为89%乙醇(V/V),液料比21∶1(V∶W),提取时间1h,提取温度55℃,该条件下模型预测的最大提取率为0.86%。[结论]应用响应面方法对黄酮类物质的提取条件进行优化是非常有效的。

Abstract:
[Objective] The aim was to study the maximum extraction conditions of flavonoids from cactus by ethanol. [Method] With dried cactus powder as material and ethanol as solvent, by using response surface methodology, the effects of ethanol concentration, extracting temperature, extracting time and ratio of material to solvent were studied, and then the regression model was established to study the extracting condition. [Result] The result of fractional factorial design indicated that ethanol concentration and ratio of material to solvent played important roles in flavonoids extracting. The regression model of extraction rate(Y) to ethanol concentration(A) and ratio of material to solvent(B) was Y=-13.587 2+0.296 1 A+0.120 3 B-0.001 74 A2-0.003 0 B2(R2 =0.948), and the optimum conditions were 89% ethanol, ratio of solvent to material 21∶1, time 1 h, and temperature 55 ℃. The maximum extraction rate predicted by the model was 0.860%. [Conclusion] Response surface methodology was a good method for optimizing extraction conditions of flavonoids.
