| 摘 要： | 本研究对已整合外源rolB-pttGA20ox双价基因的毛白杨株系进行半定量RT-PCR检测,并对移栽后的转基因植株进行形态观测。结果表明:rolB和pttGA20ox基因的表达模式不同,rolB基因在不同器官中的表达强度为根>茎>叶,pttGA20ox基因的表达无组织特异性;rolB基因受外源生长素短时间诱导时表达丰度发生变化,pttGA20ox基因表达丰度不受外源生长素短时间诱导。对移栽温室后的转化植株的生长量分析结果表明,4个转化株系及对照的高生长增加量表现为RG-4>RG-1>RG-3>RG-2>CK。4个转化株系的株高和茎间长明显高于对照。转化株系与对照之间以及转化植株之间的形态特征出现了差异。

| Abstract: | In this study,the expression of rolB-pttGA20ox double genes in transgenic lines of Populus tomentosa was analyzed by RT-PCR.The results showed the differences between rolB and pttGA20ox gene.The expression levels of rolB gene in different tissues were root>stem>leaf,and pttGA20ox gene had same expression levels in these tissues.The rolB message level was changed among exposure to the auxin treatment in 20 h and pttGA20ox gene wasn't induced by auxin treatment.Growth analysis showed that the increases of gro... |
