| 摘 要： | 为提高苜蓿花药培养效率,以小孢子发育处于单核靠边期的苜蓿花药为材料,研究了培养方法、花药接种密度以及谷氨酰胺对苜蓿花药愈伤组织诱导的影响。结果表明,采用50 mL三角瓶为培养容器,苜蓿花药采用固液双层培养,愈伤组织的诱导率最高,为33.11%;花药愈伤组织诱导率以接种密度为每50 mL三角瓶接种45枚时最高,为28.44%;培养基中附加谷氨酰胺或水解酪蛋白有利于苜蓿花药愈伤组织的诱导,其中谷氨酰胺浓度为700 mg/L时效果较佳。
