Abstract:The immune-enhancing effect of mindin was studied.The Mus musculus spon2 gene encoding mindin was synthesized with restriction sites(BamHI、HindⅢ) at 5′and 3′end respectively.The gene fragment was cloned into the expression vector Pet-22b and the HIS-tagged recombinant protein was generated and confirmed with Western blot.Mice were immunized with Sjc23 of Schistosoma japonicum in-combination with HIS-mindin.Specific to antibody responses to Sjc23 in serum was studied by ELISA.The results showed that recombinant mindin can be generated with E.coli BL21(DE3) cell lines.mindin had similar adjuvancity as the classical Freund adjuvant with respect to antibody responses to the Sjc23 antigen of S.japonicum.Thus the study provided the basis for further investigation on the application of mindin as an immune-enhancing agent in vaccine development.
