普通小麦抗条锈新种质——WT212的细胞遗传学分析及其抗性基因的RAPD标记

摘    要:对普通小麦抗条锈新种质--WT212的细胞遗传学及其抗性基因的RAPD标记进行了研究。结 果表明,WT212所携带的抗源不同于1BL/1RS易位系为基础的"洛类"抗源,而是一种来自黑麦染色体组的抗条 锈新抗源,初步断定WT212是只涉及1对染色体的小麦-黑麦易位系;RAPD分析筛选出重复性强、在抗病亲本 和抗性基因池稳定出现的特异DNA片段2个,即引物S369的扩增片段和引物S1397的扩增片段,其长度分别约 为770和1400 bp。对引物S369扩增出的特异片段与目的基因的遗传连锁性进行了分析,结果表明,引物S369扩 增出的特异DNA片段与目的基因紧密连锁。

Abstract:Wheat new germpasm WT212 with stripe rust resistance was studies by cytogenetics and RAPD techniques. The cytogenetic study result showed that the source of stripe rust resistance of WT212 is one new source from secale chromosome, different from "luolei" source which is base on 1BL/1RS translocation line. And preliminary result concluded that WT212 may be a new wheat-secale translocation line related to one pair of chromosomes. 2 additional DNA fragments were amplified repeatedly and steadily in resistant parent and resistance gene pool by RAPD technique,that is the fragments amplified respectively by the primer S369 and S1397. The additional DNA fragment amplified by the primer S369 was about 770 bp,and the fragment by S1397 was about 1 400 bp. The genetic linkage of the additional DNA fragment by the primer S369 with WT212 was tested on a segregating F2 population derived from a cross between the gene donor parent WT212 and susceptible variety Huixianhong. The result showed that the additional DNA fragment was closely linked to the target gene in WT212.
