摘    要:试验旨在探究Notch2在牛黄体化颗粒细胞中的调控机制。将免疫荧光鉴定采集到的原代颗粒细胞,通过慢病毒过表达技术感染牛黄体化颗粒细胞,最后应用qRT-PCR技术检测周期基因、抗氧化基因和孕酮合成相关基因的表达。结果显示:牛黄体化颗粒细胞过表达Notch2胞内功能片段NICD2后,周期基因CyclinD1、CyclinD2、CDK4和PCNA的mRNA表达量上升(P<0.05),而CDK1基因的mRNA表达无显著差异;抗氧化基因SOD和CAT的mRNA表达量上升(P<0.05);孕酮合成相关基因CYP11A1、STAR和3β-HSD的mRNA表达量上升(P<0.05)。综上所述,推测Notch2在牛卵泡颗粒细胞黄体化过程中促进细胞增殖、抗氧化水平和孕酮的合成。

Regulation of Notch2 in Bovine Luteinized Granulosa Cells

Abstract:The study aims to explore the regulatory mechanism of Notch2 in bovine luteinized granulosa cells.The collected primary granulosa cells were identified by immunofluorescence,the bovine luteinized granulosa cells were infected by lentivirus overexpression technique,the expression of cycle genes,antioxidant genes and progesterone synthesis related genes were detected by qRT-PCR finally.The results showed that after the overexpression of Notch2 intracellular functional fragment NICD2 in bovine luteinized granulosa cells,the mRNA expression of CyclinD1,CyclinD2,CDK4 and PCNA were increased(P<0.05),and there was no significant difference in the mRNA expression of CDK1;the mRNA expression of antioxidant genes SOD and CAT increased(P<0.05);the mRNA expression of progesterone synthesis related genes CYP11A1,STAR and 3β-HSD increased(P<0.05).In conclusion,it is speculated that Notch2 can promote cell proliferation,antioxidant level and progesterone synthesis during the luteinized process of bovine follicular granulosa cells.
