栾树离体幼胚成苗技术研究

摘    要:通过栾树离体幼胚可以产生愈伤组织和再生植株。外植体消毒时,最适宜的方法是先去种皮再消毒,消毒时间酒精处理30 s,升汞处理9 min;用1 mg·L-1的GA3对外植体进行预处理,可以促进幼胚的无菌萌发;6BA有利于幼胚的快速成苗,最佳激素浓度为2 mg·L-1,2,4D有利于产生愈伤组织,最佳激素浓度为2 mg·L-1,5 mg·L-1的6BA与2,4D易导致外植体褐化、坏死;在使用AC时,要考虑到AC的无选择吸附性,应加大使用附加物的浓度或降低AC的使用量。

Abstract:Callus and plantlets could be regenerated by culture in vitro of immature embryo of Koelreuteria paniullata . The best method for explant disinfection was to get rid of the seed coat first, and the disinfectant time would be 30 s with 75% alcohol,then 9 s with HgCl2.A pretreatment to the prerequisite with 1 mg·L-1 GA3 could accelerate the aseptic germination of the embryo, and the optimal concentration of the hormone was 2 mg·L-1;2,4D was favorable to the generation of callus, and the optimal concentration of the hormone was 2 mg·L-1.Excessively high concentrations of 6BA and 2,4D would lead to the russet and death of the prerequisite. When AC was applied, the nonselective absorption must be considered, either increase the concentration of appendix substances or decrease the dosage of AC.
