高纯度茄尼醇的分离及测定

摘    要:探讨以烟叶浸膏为原料,经醇碱溶液皂化,采用乙醇水溶液和石油醚混合溶液萃取皂化物中的茄尼醇,再以甲醇分离茄尼醇,并经层析柱纯化,制备高纯度茄尼醇;以反相高效液相色谱法(RP-HPLC)测定样品中茄尼醇含量,流动相为乙腈∶异丙醇=7∶3,检测波长为210 nm,流速为1.0 m l/m in,柱温为室温,测定结果是样品中茄尼醇含量达98.51%,平均加样回收率为98.35%,标准偏差为1.75%。

Abstract:Taking the tobacco extracts as the material,after saponified by alcohol alkaline solution,the mixture of aqueous alcohol solution and petroleum ether was use to extract solanesol from saponated substance.After isolated by methanol and purified by column chromatography,high purity solanesol was prepared.Solanesol content in samples was detected by RP-HPLC.The mobile phase was acetonitrile∶ isopropyl alcohol=7∶ 3 with 210 nm wavelength and 1.0 ml/min flow rate.The column temperature was room temperature.The experimental results showed that solanesol content from products reached 98.51%.Its average recovery rate was 98.19% and the standard deviation was 1.75%.
