棉花子叶离体培养与植株再生

摘    要:以棉花无菌苗子叶为外植体,就激素对其不定芽的分化及植株再生的影响进行了研究。实验结果表明,BA对子叶不定芽的诱导效果最好;TDZ次之,ZT和KT不能诱导子叶形成不定芽,IAA与BA配合使用,叶片不定芽分化频率明显提高;NAA与BA配合使用叶片不定芽分化频率有所下降;2,4-D对子叶不定芽分化有明显的抑制作用。在分化培养基中添加1.5mg/L GA3有利于芽的伸长。不定芽伸长后在不含激素的MS或MS+0.5mg/L NAA培养基上诱导生根,形成完全植株。

Abstract:Effects of hormones on differentiation of adventitious buds and plant regeneration were studied in cotyledon culture of cotton in vitro, BA was considered as the best growth regulat or for adventitious bud differentiation within 4 cytokinins.The combination of IAA and BA raised the differentiation frequency obviously.The higher differentiation frequency of adventitious buds was obtained in the medium MS+4.0 mg/L BA+0.5 mg/L IAA, while in the media with NAA and BA, the dif ferentiation frequency of adventitious buds reduced.However, 2, 4-D had the inhibition ef ect on the bud differentiation GA3 can promote the elongation of adventitious buds.The elongated shoots rooted in media MS without hormore or MS+0.5 mg/L IAA; and developed into the complete plant.
