山羊UBA52基因多态性及其与繁殖性能的关系

摘    要:根据牛UBA52基因序列,设计3对引物(U1~U3),分别扩增随机选取的济宁青山羊和辽宁绒山羊样本,PCR产物克隆测序,序列比对结果显示仅在U3的扩增产物中发现G63A和T181C突变。设计引物U4,利用PCR-RFLP技术分别在性早熟、高繁殖力品种(济宁青山羊、南江黄羊、马头山羊)和性晚熟、低繁殖力品种(辽宁绒山羊、内蒙古绒山羊、太行山羊)中检测上述位点的多态性,并分析2个突变位点对山羊性早熟和高繁殖力的影响。对于G63A位点,在济宁青山羊和南江黄羊中检测到GG、GA和AA3种基因型,在马头山羊、辽宁绒山羊、内蒙古绒山羊、太行山羊中只检测到GG和GA基因型;该突变位点不同基因型分布在性早熟品种和性晚熟品种间存在一定的差异;济宁青山羊GG、GA和AA基因型频率分别为0.53、0.44和0.03,AA型和GA型母羊平均产羔数分别比GG型多0.85只(P0.05)和0.49只(P0.05),AA型与GA型之间差异不显著(P0.05)。对于T181C位点,在南江黄羊和内蒙古绒山羊中检测到CC、CT和TT 3种基因型,在济宁青山羊、马头山羊、辽宁绒山羊、太行山羊中只检测到CC和CT两种基因型;该突变位点不同基因型分布在性早熟品种和性晚熟品种间没有明显差异;济宁青山羊CC和CT基因型频率分别为0.91和0.09,两种基因型个体间产羔数差异不显著(P0.05)。本研究结果初步表明,UBA52基因63位点与山羊的性早熟可能存在一定关联,A等位基因可能是提高山羊产羔数的一个潜在有效的DNA标记。

Abstract:According to Bos taurus UBA52 gene sequence,three pairs of primers(U1 to U3) were designed to clone goat UBA52 and scan polymorphisms and one pair of primers to detect polymorphisms of UBA52 in sex-ual precocious and high fecundity goat breeds(Jining Grey,Nanjiang Brown and Matou goats) and sexual late-maturing and low fecundity goat breeds(Liaoning Cashmere,Inner Mongolia Cashmere and Taihang goats) by PCR-RFLP.The results showed that there were only two base changes(G63A and T181C) in the product Cashmere and Taihang goats.There were certain differences in genotype distribution between the sexual preco-cious and sexual late-maturing goat breeds.In Jining Grey goats,the frequency of genotypes GG,GA and AA was 0.53,0.44 and 0.03,respectively.And the Jining Grey goat does with genotype AA and GA had 0.85(P<0.05) and 0.49(P<0.05) kids more than those with genotype GG,respectively.No significant difference(P>0.05) was found in litter size between AA and GA genotype in Jining Grey goats.For the 181 locus,three genotypes(CC,CT and TT) were detected in Nanjiang Brown and Inner Mongolia Cashmere goats,only two genotypes(CC and CT) were in the other four goat breeds.No significant difference was found in genotype distribution between the sex-ual precocious and sexual late-maturing goat breeds.In Jining Grey goats,the frequency of genotypes CC and CT was 0.91 and 0.09,respectively.And there was no significant difference(P>0.05) in litter size between the only two genotypes CC and CT in Jining Grey goats.The present study preliminarily indicated that there may be some association between 63 locus of UBA52 gene and sexual precocity in goats,and allele A of UBA52 gene is a po-tential DNA marker for improving litter size in goats.
