| 摘 要： | 以制备的O型口蹄疫单克隆抗体为基础,结合免疫金技术,采用柠檬酸三钠还原法制备胶体金颗粒,对6H4单克隆抗体进行标记,制备了该单克隆抗体的金标试纸条.研究结果表明,6H4单克隆抗体在pH 8.64下的稳定点为12 mg/L,可制备成金标记抗体及其反应膜. |

| Abstract: | Based on prepared monoclonal antibody(McAb) against the type O foot and mouth disease virus(FMDV),the 6H4 strain of McAb was applied to develop a new kind of McAb colloidal gold labelling strips by immune colloidal gold technique,aiming at obtaining a extraodinary,handy,rapid and audiovisual way of foot and mouth disease(FMD) detection and diagnosis.The colloidal gold particles were synthesized were prepared by sodium citrate reduction method and used as the labelling subjects of the 6H4 McAb.The results showed that the 6H4 strain of McAb was able to prepare the colloidal gold labelling antibody and its reactive membrane,and the concenration stable point of its McAb purified protein in the pH8.64 colloidal gold solution was calculated to be 12 mg/L. |
