摘    要:目的]探讨不同提取液对大叶黄杨叶绿素提取效率的影响,以及大叶黄杨叶绿素的稳定性。方法]以大叶黄杨叶片作为研究对象,比较95%乙醇、80%丙酮和95%的乙醇和丙酮混合液(体积比为1∶2)3种提取液的提取效率;探讨光照、pH值、温度、氧化剂、重金属以及紫外线对叶绿素稳定性的影响。结果]以95%的乙醇和丙酮混合液作为提取液时,大叶黄杨叶绿素的提取效果最好;大叶黄杨叶绿素的光稳定性较差;在pH值中性条件下相对稳定;具有一定的耐热性和抗氧化能力;除Cu2+使大叶黄杨叶绿素的稳定性提高外,其他重金属离子均使叶绿素稳定性降低;紫外线可使大叶黄杨叶绿素稳定性降低。结论]体积分数为95%的乙醇和丙酮(体积比为1∶2)混合液对大叶黄杨叶绿素提取效率最高,并且提取液应在避光、中性、远离重金属离子条件下保存。

Extraction and Stability of Chlorophyll in Euonymus japonicus

Abstract:Objective] To compare different extracting liquids on the extraction efficiency of chlorophyll in Euonymus japonicus,and explore the chlorophyll stability.Method] Euonymus japonicus was used as the investigate object.The effects of three different extracting liquids,namly,95% ethanol,80% acetone and 95% acetone/ethanol mixed liquor(V∶V=1∶2) on the extraction efficiency of chlorophyll in Euonymus japonicus and the effects of light,pH value,heat,oxidant,heavy metal ions and ultraviolet light on its chlorophyll stability were studied.Result] The results showed that 95% acetone/ethanol mixed liquor(V∶V=1∶2) was the best extracting liquid,and the chlorophyll of Euonymus japonicus had a poor photostability.It was relatively stable in the neutral condition,with a certain degree of heat resistance and anti-oxidant capacity.The copper ions can increase the stability of chlorophyll,while other heavy metal ions reduced the stability of chlorophyll.Ultraviolet light can reduce the stability of chlorophyll.Conclusion] 95% acetone/ethanol mixed liquor(V∶V=1∶2) has the best extraction efficiency of chlorophyll.The extracting liquid should keep in dark,neutral condition without heavy metal ions.
