摘    要:为了建立一个高效的金达苜蓿(Medicago sativa L. cv. Jinda)组织培养再生体系,我们以金达苜蓿无菌苗的胚轴上段、下段、子叶为外植体,进行了2,4-D和6-BA不同组合配比对愈伤组织诱导和胚性愈伤组织及胚状体分化的影响,这对其以后建立遗传转化体系具有重要的意义。结果表明:胚轴上、下段和子叶是很好的诱导愈伤组织的外植体;在MSO胚状体诱导培养基上,分化出胚性愈伤组织的最佳2,4-D、6-BA剂量配比为8:0.2,成苗培养基为MSO+30 g/L蔗糖+6 g/L琼脂。

Abstract:The upside and downside of hypocotyls and cotyledon of Medicago Sativa L.cv.Jinda were used to study the effect of different concentrations of 2,4-D and 6-BA on the callus induction and embryogenic callus, embryoid differentiation. The results showed that the upside and downside of hypocotyls, cotyledon were good explants for Jinda. The best combinations and concentrations of 2,4-D、6-BA is 8/0.2 in inducing embryoid medium MSO. The best medium for plant development was MSO+30 g/L sucrose + 6 g/L agar. The goal of this research is to establish an effective regeneration system of Medicago Sativa L.cv.Jinda.by tissue culture and it has important significance for establishing genetic transformation system.
