中药复方宫炎净不同溶剂提取物对小鼠耳廓肿胀及毛细血管通透性的影响

摘    要:通过研究中药复方宫炎净不同溶剂提取物对小鼠耳廓肿胀和毛细血管通透性的影响,筛选抗炎作用最好的溶剂提取物。将110只21日龄雌性小鼠随机分为11组,每组10只,分别给予生理盐水,地塞米松及高、中、低剂量的石油醚提取物、乙醇提取物、水提取物,连续灌胃4d后,用二甲苯致炎,测定小鼠耳廓肿胀率。此外,另取110只小鼠按上述方法分组、给药,并在最后1次给药后腹腔注射体积分数为0.7%的冰醋酸致炎,于590nm波长处测腹腔液OD值。结果显示,乙醇提取物各剂量组的耳廓肿胀率显著低于生理盐水组和石油醚组(P0.01),并以乙醇中剂量提取物效果最佳;乙醇提取物各组和地塞米松组的腹腔毛细血管通透性均极显著低于除地塞米松组外的各组(P0.01),并且乙醇中剂量组显著低于地塞米松组(P0.05)。说明,乙醇可作为提取该复方有效成分的最佳提取溶剂。

Abstract:In order to screen the solvent extracts with best anti inflammatory effects, different solvent extracts of traditional Chinese medicine compound Gongyanjing were used to study their effects on mouse ear edema and capillary permeability. 110 21 day old female mice were randomly divided into 11 groups, ten for each group. Treatments of saline, dexamethasone, three dosages (high, medium and low) of petroleum ether extracts, ethanol extracts and water extracts were given by gavage for four days, then with the xylene induced inflammation, mouse ear swelling rate was measured. In addition, an alternative 110 mice were grouped and treated with same method. After four days treatment, inflammation was induced by intraperitoneal injection of 0.7% acetic acid, and OD value of peritoneal fluid was measured at wavelength of 590nm.The results showed that ear swelling of ethanol extracts treatment groups were significantly lower than that of the saline group and petroleum ether group (P<0.01), and the effect of ethanol extracts with medium dose was the best; Peritoneal capillary permeability of ethanol extracts and dexamethasone treatment groups were significantly smaller than other groups (P<0.01), and medium dose ethanol extracts group was significantly lower than that in the dexamethasone group (P<0.05). As a conclusion ethanol was the best extraction solvent for extracting the active ingredient from the compound.
